This gene, part of the PDGF/VEGF family, encodes a heparin-binding, disulfide-linked homodimeric growth factor essential for angiogenesis. It promotes vascular endothelial cell proliferation and migration, with gene disruption in mice causing abnormal embryonic vessel formation. Upregulated in many tumors, its expression correlates with tumor progression. High protein levels are seen in POEMS syndrome and associated with diabetes-related microvascular issues and atherosclerosis. Multiple isoforms arise via alternative splicing, non-AUG initiation, and stop codon readthrough, with one extended isoform being antiangiogenic. Some AUG-derived isoforms are regulated by a uORF within an IRES. VEGF levels increase in SARS-CoV-2 infection, promoting inflammation via inflammatory cell recruitment and upregulation of angiopoietin II, creating a feedback loop that amplifies cytokine release.
